<A. Lambert> is the first author of a 2000 study improving measurements of <follicle-stimulating hormone> effects on <aromatase> in human <granulosa-lutein cells>. These <cells>, collected during in-vitro fertilization procedures, convert <testosterone> to <estradiol> through <aromatase>. The study addressed a measurement problem: hormone output during a long incubation depends both on the amount of active <enzyme> and on how much <substrate> remains.<Improved FSH sensitisation and aromatase assay in human granulosa-lutein cells by A. Lambert and others>[Abstract: human follicular aspirates and interpretation of prolonged testosterone exposure]{ref}

The researchers separated the stages. They first preincubated the <granulosa-lutein cells>, stimulated them with <follicle-stimulating hormone> for 24 hours, washed them, and then supplied <testosterone> for a two-hour assay. The short challenge limited <substrate> depletion and isolated the later measurement from the earlier stimulation. After preincubation, the observed stimulation was three- to fourteen-fold over basal activity, depending on the preparation. This illustrates why an <enzyme> assay needs control of both biological regulation and reaction conditions.<Improved FSH sensitisation and aromatase assay in human granulosa-lutein cells by A.
