= Formazan optical response
A \b[formazan optical response] is an observed optical signal associated with a specified <formazan> derivative and its physical/chemical state. It can depend on dissolved product concentration, wavelength, solvent, optical geometry and suspended material. The family name alone does not supply one universal <molar absorption coefficient>.
For a suitable dissolved-product measurement, the <Beer-Lambert law> gives
$$
A-A_0=\epsilon\ell c_F=\epsilon\ell\frac{n_F}{V}.
$$
Here $A$ is <absorbance>, $A_0$ the relevant background, $\ell$ the geometric light path, $n_F$ dissolved amount and $V$ solution volume, with compatible units. In a fixed-path cuvette, doubling volume at fixed amount halves the ideal product absorbance.
A vertical measurement through an ideal parallel-sided well behaves differently. If well area is $S$, fill height is $\ell=V/S$, so
$$
A-A_0=\epsilon\frac{n_F}{S}.
$$
This <microplate fill-height optical cancellation> assumes the same represented area and optical geometry. Dilution lowers concentration while increasing path length, so the volume factors cancel. A real well's meniscus, beam geometry and instrument processing can require a more detailed model; this is not a universal correction for every plate reader.
A controlled dilution check should therefore specify whether path length is fixed or changes with fill height. In an original fixed-path example, product contribution $1.2$ plus a constant optical background $0.4$ gives raw absorbance $1.6$. Halving product concentration with that same background gives $1.0$, not half the initial raw value. Subtracting the correct background recovers product contributions $1.2$ and $0.6$.
The manufacturer's <MTT> principle separates formation of formazan crystals from their later dissolution and multiwell measurement.<Cell Proliferation Kit I MTT instructions by Roche>[Section 3.1 and Figure 4, printed p. 9]{ref} Visible particles can introduce light scattering, which is not automatically described by a dissolved-dye absorption law. An <MTT formazan solubilization check> addresses this physical stage.
A spectrum or a single absorbance value is an optical observation. Inferring <cell viability> additionally requires a validated biochemical response model, because <tetrazolium-reduction interference> can produce the same colored material without the intended cellular mechanism.
