Heparosan synthase (source code)

= Heparosan synthase

<Heparosan synthase> is an <enzyme> activity that assembles the alternating <glucuronic acid> and <N-acetylglucosamine> backbone of <heparosan>. Its activated donors include <UDP-glucuronic acid> and <UDP-N-acetylglucosamine>. Backbone synthesis is distinct from subsequent modification such as <N-sulfation of glucosamine> or <epimerization>.<Identification and molecular cloning of a heparosan synthase from Pasteurella multocida type D by P. L. DeAngelis and Carissa L. White>[Abstract, donor pair and nonsulfated, unepimerized polymer product]{ref}

The 2002 study of <Pasteurella multocida> type D cloned a 617-residue <protein> and tested a recombinant preparation produced in <Escherichia coli>. Incorporation from activated sugars labeled with <radioisotopes> supplied evidence for polymer formation; detecting the recombinant <protein> alone would establish expression, rather than the claimed chain-building reaction.<Identification and molecular cloning of a heparosan synthase from Pasteurella multocida type D by P. L. DeAngelis and Carissa L. White>[Abstract and Activity Assays, recombinant preparation and radiolabeled donor incorporation]{ref}

A constructed donor-limitation model illustrates the balance. Assume a supplied acceptor population, no chain initiation or donor side reactions, and equal numbers of additions of the two sugars. Starting with 30 nanomoles of <UDP-glucuronic acid> and 20 nanomoles of <UDP-N-acetylglucosamine> permits at most $\min(30,20)=20$ nanomoles of complete two-sugar repeat equivalents. That corresponds to 40 nanomoles of incorporated residues and leaves 10 nanomoles of the first donor when only these balanced additions are counted.

This ceiling concerns total incorporation, not the number of polymer molecules or their individual lengths. Terminal residues can also make real chains depart from exact whole-repeat accounting. Measuring product size and chemical identity alongside incorporation therefore distinguishes more possibilities than a single bulk signal. The characterized bacterial <enzyme> supplies a concrete implementation; it does not establish that every organism builds or modifies related polysaccharides with the same protein architecture.
