P element (source code)

= P element
{c}

A <P element> is a <DNA transposon> studied in <Drosophila melanogaster>. Its movement depends on recognition of the element's ends and the activity of <P-element transposase>. This separates two requirements: the mobile <DNA> needs appropriate recognition sequences, while the enzyme enabling movement can be supplied from another source.<P element in FlyBase>[Natural-transposon record, transformation-vector and transposase-source sections]{ref}

That separation made the <P element> useful for <genetic transformation>. In1982, investigators inserted a wild-type <Drosophila melanogaster rosy> sequence into a <P-element vector> and introduced it into mutant embryos. The resulting germline transformants carried chromosomally integrated copies that were inherited in later generations and restored the eye-colour phenotype. Transformation occurred in20–50percent of the injected mutant embryos in that experiment.<Genetic transformation of Drosophila with transposable element vectors by G. M. Rubin and A. C. Spradling>[Abstract, ry1 construction, inherited copies and eye phenotype]{ref}

The observations answer different questions. A restored visible trait shows that the introduced <gene> can function in its new context; chromosomal analysis and transmission to descendants establish integration and inheritance. Neither observation alone identifies every insertion site or ensures that another inserted sequence will have identical expression. The <P element> is therefore both a biological mobile element and a basis for experimental tools whose outcomes depend on construct and genomic context.
