Use <RNA sequencing> and quantify reads spanning the exon-4/exon-5 and exon-4/exon-6 <splice junctions>, with appropriate normalization for informative read opportunities. These distinguish production of the long- and short-associated <transcript isoforms> rather than merely measuring total <TCF transcription factor> transcripts. Adequate junction coverage and biological replication are required; <long-read RNA sequencing> can independently check whether the assumed complete isoforms are present. This avoids relying on a comparison of raw Ct values from two different <PCR primer> pairs.
