A 1991 collaborative paper provides a specific contribution to understanding <transfer RNA> identity. Varshney and colleagues altered an <initiator transfer RNA> in <Escherichia coli> and tested its function in both initiation and chain elongation. Internal amber <stop codon> suppression measured elongator activity; one variant could also initiate at an upstream UAG and suppress a downstream UAG. This distinguishes two questions that an amino-acid label alone cannot answer: what chemical cargo a <transfer RNA> carries, and which step of <biological translation> recognizes that carrier. Testing the same RNA at an initiating position and an internal <codon> probes a functional distinction, rather than merely comparing two RNA sequences.
