There is no large, consistent stimulation by added Wnt protein ligand in the displayed experiment. The paired bars are close in the empty-vector and TCF-long conditions; TCF-short shows a modest upward difference, while some other pairs decrease slightly. The error bars and absence of raw replicate information do not permit a firm significance claim. This result concerns the particular reporter and transfection conditions, not every aspect of endogenous Wnt signalling. A known ligand-responsive positive control, dose/time testing and validation of receptor/pathway competence would distinguish a genuinely weak response from failed stimulation or saturation caused by transgene expression.
