Let G count copies of allele 2. Under Hardy-Weinberg equilibrium, G has a binomial distribution with parameters 2,ϕ, and the penetrance is qG=pθG. Consequently
Pr(Y=1)=p(1−ϕ)2+θp2ϕ(1−ϕ)+θ2pϕ2=p[(1−ϕ)+ϕθ]2=pα2.
(1)
Thus the overall penetrance is
K=pα2,α=1+ϕ(θ−1).
(2)
The multiplicative diallelic penetrance model requires 0≤ϕ≤1, p≥0, θ≥0, and each occupied genotype's value pθG to be at most one. These are probabilities, not unrestricted relative-risk scores.
