Homotrimer 2026-11-05
A homotrimer is a protein trimer containing three copies of the same protein subunit type. “Homo” specifies composition; “trimer” specifies a total of three subunits. A homotrimer need not have perfectly identical instantaneous conformations or ligand occupancies at all three copies.
Polynucleotide phosphorylase provides a functional example. Its three repeated subunits form a ring around the polynucleotide phosphorylase central channel. Cryo-electron microscopy of the mycobacterial enzyme resolved that ring and RNA near a catalytic site, connecting an oligomeric structure with an RNA-processing mechanism.[ref]
For an original conservation example, let be an unassembled chain and the assembled homotrimer, with no other forms present:
A total chain concentration of can yield at most trimers. If half the chains remain unassembled, only trimers form. Reporting concentration in chains or in complexes therefore changes the numerical value even for the same preparation.
Mihaela-Carmen Unciuleac 2026-11-05
Mihaela-Carmen Unciuleac is a joint first author of a 2021 study connecting polynucleotide phosphorylase structure with its reversible reactions at an RNA chain's end. In one direction, phosphorolysis releases nucleoside diphosphates; in the other, those substrates supply units for extension.[ref]
The study combined cryo-electron microscopy with changes to selected amino acid residues and measurements of enzyme activity. The structure localized substrate and metal-binding features; the activity comparisons tested their functional importance.[ref]
For an original accounting example, shortening an RNA from 12 to eight nucleotides through four terminal phosphorolysis events consumes four inorganic phosphate equivalents and releases four nucleoside diphosphates. Reversing those four events consumes four nucleoside diphosphates and releases four phosphate equivalents. This balances the two directions without assuming that a reaction mixture must run equally fast both ways.
A structural contact also differs from a demonstrated chemical requirement. In the paper's Figure4, an imported comparison phosphate helps interpret the active site; it is not a directly resolved phosphate from that same reconstruction.[ref] Comparing structural placement with separate reaction measurements makes the mechanistic claim testable without assigning unreported individual tasks to an author.
Mycobacterium smegmatis 2026-11-05
Mycobacterium smegmatis is a bacterial species used as an experimental model for mycobacterial molecular biology. Species identity and laboratory strain identity are different levels of description: the ATCC record for the genome-sequenced strain identifies a particular lineage, not every member of the species.[ref]
Its polynucleotide phosphorylase illustrates the model's scientific value. Researchers combined cryo-electron microscopy with enzyme assays to locate RNA in a three-subunit ring and test the contributions of selected amino acid residues. Comparing structure with catalytic measurements connects a molecular arrangement to a biochemical function.[ref]
For an original interpretation example, suppose a purified reference enzyme gives100 activity units and a substituted variant gives25 under the same stipulated assay conditions. The variant retains of that measured activity. This is evidence about the tested preparation and reaction; it does not by itself establish a fourfold change in bacterial growth or the same effect in Mycobacterium tuberculosis.
A useful model permits controlled comparisons, but its value does not make distinct species, strains or experimental readouts interchangeable. Reporting the strain and molecular system keeps the inference tied to what was actually measured.
The polynucleotide phosphorylase central channel is the passage through the ring-shaped protein trimer formed by polynucleotide phosphorylase. It helps organize RNA access to catalytic regions of the assembly. The channel is a property of the assembled enzyme, rather than a hollow tube inside one isolated polypeptide.[ref]
Different experiments supply different levels of evidence. The 2000 structure contained a phosphate analogue but no bound RNA and proposed a channel role in processivity. A 2021 cryo-electron microscopy study captured an RNA-containing state of a mycobacterial enzyme, including a terminal oligonucleotide in an active site and an adenine base in the central tunnel. That directly places substrate features in the assembly without resolving every segment of a complete chain.[ref]
A conceptual geometry test illustrates the importance of routing. Imagine an RNA with a flexible single-stranded tail followed by a bulky paired region. A channel admitting the tail need not admit that paired region. Entry, retention and bond cleavage can therefore impose different constraints on the observed reaction, even when the chemical active site is unchanged.
The structural observation should not be interpreted as a molecular movie. Mobility, unresolved density and transitions between catalytic states require biochemical or kinetic evidence alongside the static structure. In particular, a ring-shaped assembly alone does not prove unlimited processivity.
Shreya Ghosh 2026-11-05
Shreya Ghosh is a joint first author, with Mihaela-Carmen Unciuleac, of a 2021 structural and biochemical study of polynucleotide phosphorylase from Mycobacterium smegmatis. The enzyme links reversible RNA end chemistry with the architecture of a ring-shaped protein trimer.[ref]
The researchers used a Titan Krios G2 transmission electron microscope with a Gatan K3 camera for cryo-electron microscopy. Their reconstruction located a terminal RNA segment and two magnesium ions in an active site, and an adenine base in the polynucleotide phosphorylase central channel.[ref]
A useful original interpretation test is to compare a structural map with a reaction time course. Suppose a map resolves four terminal nucleotides, while an independent assay measures conversion of a 12-nucleotide chain to an eight-nucleotide chain. The map identifies an occupied configuration; the assay establishes net shortening by four units. Neither alone establishes how long each intermediate persisted. Combining positional and functional evidence answers more than treating a structural snapshot as a movie.
The paper's joint authorship establishes a shared scientific contribution. It does not assign each instrument operation or analysis step to Ghosh individually.
