During the epoch with j ancestral lineages, Kingman's coalescent has waiting time Tj∼Exp((2j)). In the stated time units, each lineage accumulates mutations at rate Nu=θ/2, so the mutation count on a neutral coalescent tree decomposes as
S=∑j=2nYj,Yj∣Tj∼Poisson(2jθTj).
(1)
The law of total expectation and the mean of an exponential distribution give EYj=(jθ/2)/(2j)=θ/(j−1). Therefore
ES=θHn−1,Hn−1=i=1∑n−1i1.
(2)
This calculation counts mutation events. Equating the count with observed segregating sites additionally uses the infinite sites mutation model.
