Factor Va 2026-11-05
Factor Va is the activated form of Factor V that serves as a protein cofactor for factor Xa during prothrombin activation. In the membrane-associated complex, factor Xa supplies the proteolytic catalyst while factor Va helps organize efficient conversion to thrombin.[ref]
Crystal Structure of Prothrombin Reveals Conformational Flexibility and Mechanism of Activation by Nicola Pozzi and others. Introduction: prothrombinase components and substrate interactions
The cofactor's effect can be measured by reconstituting the reaction from purified components. In a bovine study with one defined phospholipid mixture, adding factor Va reduced the apparent Michaelis constant for prothrombin from to . Other vesicle compositions gave different relationships between binding and apparent kinetics.[ref]

The ratio describes the change in that apparent kinetic parameter. It does not imply that every reaction rate rises 55-fold: the observed rate also depends on substrate abundance and the limiting rate. The example shows how a noncatalytic protein can strongly regulate an enzyme reaction by participating in its molecular assembly.
Nicola Pozzi 2026-11-05
Nicola Pozzi investigates the molecular mechanisms of thrombosis, including how protein structure controls the conversion of prothrombin into thrombin. His work combines biochemical measurements with structural and single-molecule methods.[ref]
In a 2013 study, Pozzi and colleagues combined X-ray crystallography with measurements of distances within prothrombin in solution. Their engineered protein lacked the Gla domain and replaced catalytic serine 525 with alanine. Measurements supported two major arrangements in solution rather than one rigid shape. The distinction matters because prothrombin is a zymogen: activation requires cutting particular peptide bonds, and folding can expose or conceal the relevant sites. The study connected structural flexibility with access to those sites; its engineered construct and experimental conditions limit how directly a single observed arrangement can describe native prothrombin in every setting.[ref]

A zymogen is a precursor of an enzyme that becomes substantially more active after a chemical activation step, commonly limited proteolysis. Regulating activation permits a biological cell or organism to produce a catalytic precursor before the full activity is needed. Low precursor activity need not be exactly zero in every assay.
Prothrombin provides a concrete example. Specific cleavage sites connect its precursor architecture to generation of thrombin. A 2013 structural study examined an engineered precursor lacking one domain and carrying an inactive catalytic-site substitution, supplemented by solution measurements. It related exposure of activation sites to conformational organization.[ref]

A zymogen can therefore be a folded, regulated protein rather than an unfinished chain awaiting protein folding.Consider a synthetic assay in which 20 percent of a precursor pool becomes active enzyme, one molecule for one molecule. Active enzyme is then . At saturating substrate and an illustrative turnover number of , the predicted initial reaction rate is . Measuring total precursor-plus-product abundance would not reveal that active fraction.
