Sanger sequencing controls 2026-10-18
The endpoint argument identifies what its scientific controls must interrogate. A known-template comparison tests whether preparation, copying and detection can produce an interpretable ladder. A no-template comparison tests background arising without the intended template. Agreement from an independently primed opposite-strand read tests the proposed nucleotide sequence through its reverse complement. These are distinct analytical checks; none follows merely from seeing a gel band.
An ambiguous ladder should retain its ambiguity. More than one band at a nominal position can reflect mixed templates or unwanted stops; a missing band can reflect failed detection rather than an absent nucleotide. Four clean lanes are an assumption of the ideal example. They do not establish a unique genome, a biological function, or the frequency of a mutation in a population. The historical observations below show why a control for copying chemistry matters even before interpreting the letters.
Somatic cell nuclear transfer Created 2026-10-14 Updated 2026-10-20
Somatic cell nuclear transfer introduces a body-cell cell nucleus into an ovum whose own nuclear contribution has been removed or inactivated. The donor nuclear genome and recipient cytoplasm have different origins; successful development does not show that the donor cell nucleus acts independently of the recipient's machinery.
Species 2026-10-13
A species is a named unit of biological classification. The useful criteria vary among groups: reproductive compatibility can inform classification in many animals, while microbial comparisons also use genomes and other characters. Escherichia coli is a species; K-12 MG1655 identifies a particular strain within it. See OpenStax Biology 2e, 18.2: species concepts.
Splice-aware sequence alignment 2026-10-20
A sequence alignment that permits gaps corresponding to removed introns when matching processed RNA reads to a genome. Short microexons require particular care because their supporting aligned segments can be very small.
Transposable element 2026-10-20
The hypothesis that two early large-scale duplication rounds contributed to the multiple paralogous genomic regions of jawed vertebrates. Hox clusters are one informative set of loci; a single family does not by itself establish genome-wide duplication.
