The large breakage frequencies occur mainly at supraphysiological NVNA1 levels; the probability at the mean latent level is not measured directly. At 20 ng/ml doxycycline, NVNA1 is already about twice the latent amount, and breakage is about 8%. At 50 ng/ml, the protein is approximately eightfold above latent levels and breakage about 25%; at 100 ng/ml, approximately sixteenfold and about 45–50%. Higher induction reaches roughly 60% while expression rises to about 21–24 times the latent level. The western blot loading control supports a protein-abundance comparison, although quantitative interpretation also requires unsaturated immunoblot detection.
Latent expression, defined as a ratio of one, lies below the lowest nonzero measured dose. Extrapolating a straight line from baseline and the twofold point would suggest a small frequency, but neither linearity nor a threshold is established. The justified conclusion is lower expected short-term breakage than in the strongly induced conditions, with a potentially important effect of modest upward fluctuations. Cell-to-cell variability, duration of expression and repeated divisions could make uncommon events consequential during a persistent infection. A percentage of broken chromosomes in this assay is not a patient's lifetime cancer probability. Doxycycline and tag controls are also needed to attribute the induced-cell phenotype specifically to NVNA1.
