A calibrated coordinate in a harmonic trap has equilibrium variance ⟨δx2⟩=kBT/k. This determines stiffness only after removing detector noise and accounting for drift, exposure averaging and finite bandwidth. Position variance does not supply a force calibration if the coordinate scale is unknown.
