Glucokinase regulatory protein binds and regulates hepatic glucokinase, including its sequestration in the nucleus under suitable conditions. Glucose and fructose-phosphate metabolites alter this interaction, coordinating enzyme availability with nutrient context.
Glucokinase phosphorylates glucose and has kinetic properties suited to sensing changing glucose availability in liver and pancreatic beta cells. Its activity is not strongly inhibited by glucose-6-phosphate, and tissue-specific regulation determines how that flux is used.
