Transiently binding fluorescent probes provide spatially separated single-emitter observations whose fitted positions accumulate into a structural map. Probe exchange can replenish bleached emitters. Precision, specificity, event density, optical background and drift all affect the usable resolution; accumulating events does not itself remove systematic labelling error.
Reversible complementary-strand recognition supplies transient fluorescent docking events on a DNA nanostructure. Individual events are localized and accumulated as in PAINT.[ref]
