Mei-Chu Lo investigated experimental methods for detecting ligand binding to proteins. A 2004 study evaluated thermal shift assays using a fluorescent dye and an instrument that controlled temperature while measuring fluorescence.[ref]
Evaluation of fluorescence-based thermal shift assays for hit identification in drug discovery by Mei-Chu Lo and others. Analytical Biochemistry332, pp.153–159; Abstract and primary byline
The study compared binding estimates with isothermal titration calorimetry and examined relationships with enzyme activity assays. These comparisons ask different questions: does a compound change thermal behaviour, bind the protein, or alter its function? Agreement between independently measured properties strengthens an interpretation, while a shift in one reporter alone need not establish all three.[ref]
