Permanganate-iron endpoint and blank model 2026-10-18
The apparatus and motivating measurement are described in permanganate-iron titration; the following is an original account of its possible errors. Assume correct titrant standardization, the acidic permanganate-iron balance, quantitative conversion of the sampled , and additive contributions expressed as equivalent titrant volumes. WriteHere is the sample titre, the titration blank, background reagent consumption, and signed offsets of the observed titration endpoints from the respective stoichiometry conditions after background consumption. The subscripts distinguish sample and blank. is additional sample-specific permanganate demand, absent from the blank; is apparent delivery used to fill an initially air-containing burette tip rather than enter the flask. Assume the blank's tip is already filled; the sample should also have after proper preparation. These terms are a diagnostic model, not measured components of the stipulated blank.
Subtracting the two accounts proves the error relationThe simple inference is valid when this residual is negligible. Matching water and sulfuric acid tests the background terms, but does not by itself establish : flask composition and viewing conditions can differ. A coloured sample can mask residual permanganate, changing the assigned titration endpoint without a corresponding redox consumption. A sample-specific reducing agent instead contributes positive by consuming permanganate. A reagent titration blank cannot measure either effect just by omitting the sample.
A transient purple streak before mixing is not the mixed-flask titration endpoint. Conversely, the equivalence point is the five-to-one inventory match, without a promised visible flag. In the ideal selective model, a persistent excess colour requires some permanganate beyond that match. No particular drop size, waiting time or endpoint offset is stipulated here; the same stated colour criterion must be applied to sample and blank, and its transferability checked.
The model also predicts error direction. If a tip bubble disappears during sample delivery, makes the inferred amount of substance too high because final minus initial burette reading includes liquid that stayed in the tip. Conditioning the tip before the first reading prevents this particular discrepancy; subtracting a separate titration blank is not an instrument repair. Repeated aliquots test scatter, a known added inventory tests the five-to-one response, and an independent titrant standardization tests . These are the proposed controls from permanganate-iron titration, with distinct diagnostic roles; none alone proves absence of all sample interference.
Permanganate-iron titration Created 2026-10-14 Updated 2026-10-20
A proposed modern teaching measurement puts a pipette-measured portion of an iron chemical solution in a conical flask, acidifies with excess dilute sulfuric acid, and adds standardized potassium permanganate from a clamped burette while swirling. The documented endpoint principle is that added purple permanganate is consumed until the first small excess leaves a pale purple colour; no separate colour indicator is required. Insufficient acidity can produce brown manganese dioxide, invalidating the assumed five-electron half-reaction.[ref]
Iron Tablet Determination Analytical Competition (Royal Society of Chemistry resource). “Setting the Scene”, final two paragraphs: burette, conical flask, decolourisation, endpoint and excess sulfuric acid
Record initial and final burette readings; their difference is the delivered volume . The following controls are proposed additions to test the inference. Run a reagent blank with the same water, sulfuric acid and endpoint criterion but no sample, obtaining . Use a separately established potassium permanganate concentration , clear the burette tip of air, and repeat sample portions. A known added amount of substance of should increase consumption by one fifth of that amount. A blank estimates reagent demand and the small endpoint excess; it cannot remove an interfering sample reactant.
Assuming complete, selective reaction according to the acidic permanganate-iron balance, the corrected inference isWith in , use and in litres. As an original stipulated calculation, not an experimental result, take , and . Then the permanganate consumption is and the inferred amount is . In a sample portion this corresponds to .
In this example the corrected volume is . The acidic permanganate-iron balance gives for and for permanganate. Thus of permanganate accepts of formal electrons, exactly what of supplies. The amount of substance ratio is five to one, despite equality of the exchanged electron amounts.
The denominator is the original aliquot, not its final volume after adding sulfuric acid, water and titrant. Such dilution changes concentrations in the flask without changing the initial sampled amount of substance of . Omitting the titration blank would instead give and , an overestimate of relative to the corrected constructed value. The displayed digits express the stipulated calculation, rather than establish its measurement uncertainty.
The endpoint detects residual permanganate, not the iron identity directly. Other sample reactants that consume permanganate raise the apparent amount; prior oxidation of sample to lowers it. A recovery check probes response to the added iron but does not prove absence of every interference. The readout determines the reacting inventory under the stated conditions, rather than total iron in an arbitrary mixture. No measured titres are asserted here.
Phase diagram Created 2026-09-24 Updated 2026-10-14
A phase diagram partitions a space of thermodynamic variables into regions occupied by different equilibrium thermodynamic phases and records the phase coexistence curves between them. For a pure, one-component substance, a pressure–temperature phase diagram gives one point for each pair . In a single-thermodynamic phase region both controls can vary independently. On a two-thermodynamic phase phase coexistence curve, they obey one relation; at a triple point, both are fixed. These statements assume bulk thermodynamic equilibrium, fixed composition and no additional applied fields.[ref]
Gibbs Phase Rule (MIT 10.213 course material). Gibbs Phase Rule, single-, two- and three-phase counts and final pressure–temperature diagram paragraphs
At fixed temperature and pressure, the stable bulk thermodynamic phase minimizes Gibbs free energy for a fixed amount of substance. For a pure substance this is a comparison of chemical potentials on a common normalization. Two candidate thermodynamic phases coexist where and neither is exceeded in stability by a third thermodynamic phase. Differentiating their equality gives the Clapeyron equation. Thus a phase coexistence curve is a locus of equal stability, rather than a trajectory that every heated sample must follow.[ref]
The following original cartoon uses dimensionless positive coordinates and , with arbitrary reference scales . It represents no particular substance and contains no measured data. Stipulate a triple point at , a liquid-gas critical point at , and these drawing rules:They specify sublimation, liquid–gas phase coexistence and melting boundaries for the picture. They are not an equation of state, do not predict latent heat or mass density, and do not supply a quantitative critical theory. In particular, the melting slope is merely an illustrative choice.
Generic dimensionless pressure–temperature phase diagram: curves, triple point and liquid-gas critical point
. Original cartoon with stipulated drawing rules. Three distinct bulk thermodynamic phases coexist at ; the liquid–gas boundary ends at . The fluid region beyond that endpoint is connected: no extra boundary is implied by the fading background. These coordinates are not water data. Details. For example, at fixed the melting boundary is at and the liquid–gas boundary is at . Heating from to therefore starts in liquid, crosses phase coexistence at , and ends in gas. In an equilibrium conversion at that crossing, added heat can change the phase fraction at fixed temperature through latent heat. The pressure–temperature point alone does not specify how much of each thermodynamic phase is present: for that one needs an additional amount or volume constraint.
Now connect exactly the same endpoints by raising from to at , moving to at , then lowering to . The first leg stays on the liquid side because ranges only from to and . The second leg lies above the entire liquid–gas boundary, whose largest drawn pressure coordinate is ; the third lies beyond its largest temperature coordinate, . No leg crosses a first-order phase transition boundary. The middle leg passes through a supercritical fluid region, with and , and the final leg returns to a gaslike state continuously.
The same liquid and gas endpoints: crossing phase coexistence or going around the liquid-gas critical point
. Original dimensionless cartoon. The orange constant-pressure path crosses the liquid–gas boundary at . The blue path follows to to to and avoids every drawn phase coexistence curve. Dashed coordinate guides locate the critical endpoint without representing additional boundaries. Details.Avoiding a first-order phase transition does not mean that no heat or work is exchanged. Along the detour, mass density, enthalpy and heat capacity may change substantially but continuously. The liquid-gas critical point itself has singular behavior, so the detour goes around it rather than through it. This connected fluid region is the striking lesson of a phase diagram: a familiar change from liquid to gas need not involve boiling across a sharp boundary.
An equilibrium phase diagram also does not predict how rapidly a conversion occurs. Metastability and a barrier described by classical nucleation theory can delay the observed phase transition. Nor should a binary-composition phase diagram be read as the same picture: its coexistence regions and lever rule encode composition and phase fractions, and can occupy an area even though pure-substance two-thermodynamic phase coexistence occupies a curve in the pressure–temperature plane.
Pipette Created 2026-10-14 Updated 2026-10-18
For an acid-base titration, a volumetric pipette selects an aliquot whose amount of substance is from a homogeneous sample. The delivered volume depends on the instrument's calibration and drainage convention; the residual liquid of a gravity-draining volumetric pipette is allowed for in that calibration. Its mark is read using the liquid meniscus. A volume calibration alone does not establish that the sampled chemical solution has the intended concentration.[ref]

Pollen tube germination assay 2026-10-14
A pollen tube germination assay makes tube emergence visible outside a flower. The published Practical Biology experiment, Observing the growth of pollen tubes (Practical Biology), Technical note 3, lists florist lilies among successful materials without specifying a species. Record the actual identification; success with one sample cannot establish a universal growth rate.[ref]
Observing the growth of pollen tubes (Practical Biology). Technical note 3 and Teaching notes: material dependence and contorted versus straight tubes
Pearson's Core practical 4: Investigate the effect of sucrose concentration on pollen tube growth by Pearson Education Ltd, Teacher sheet, pp. 1–2, describes an inspectable experiment. Mix equal volumes of sucrose solution and mineral-salt medium on clean cavity slides. Dust in pollen from mature anthers with a mounted needle, record the addition time, and incubate without coverslips in covered dishes containing wet filter paper. The moist chamber reduces drying; the uncovered drops avoid oxygen limitation. Brief observations with an optical microscope at , using an eyepiece graticule calibrated against a stage micrometer, reveal tube emergence and length. Return slides to the chambers and switch off the lamp between observations to reduce heating and drying. For comparisons of sucrose concentration, use the same flower across treatments to control maturity, then repeat with other flowers.[ref]
An additional proposed experimental design check is a medium-only slide: it tests whether debris is being mistaken for pollen tubes. Compare matched pollen samples at different final sucrose concentrations, keeping temperature, elapsed time and observation rules fixed. A sample known to produce pollen tubes in a previously successful medium would test whether an apparent failure comes from handling or the tested condition. These controls are questions to test, not guaranteed outcomes. A negative result cannot distinguish dead pollen from an unsuitable medium without further comparisons.
Two original calculations clarify the readout. First, take of sucrose solution and of sucrose-free medium. Assuming additive volumes, conservation of the amount of substance givesThis dilution also halves the medium's original component concentrations. Label the final molar concentration, not the starting bottle.
Second, stipulate a toy field containing pollen grains, of which have emerged pollen tubes. Its emergence fraction is . If those twelve pollen tubes have total measured length , their arithmetic mean length is . Assigning length zero to the other grains instead gives per grain. These invented numbers show why emergence fraction and mean length among emerged tubes answer different questions. Specify the denominator and the emergence criterion before counting, and avoid selecting only conspicuous long pollen tubes.
Tube emergence in a drop demonstrates growth under that medium's conditions. It observes no stigma interaction, passage through a style, delivery into an ovule, gamete fusion, or seed development. Thus a positive pollen tube germination assay does not demonstrate successful fertilization in a living flower. Its beauty is making one otherwise hidden part of sexual reproduction directly observable.[ref]

Primary standard 2026-10-18
A primary standard supplies a reliably known reacting inventory from which a titrant concentration can be determined.
The defining advantage is known high purity: the weighed portion can be assigned a known amount of substance. A titrant whose concentration is established against that inventory is a secondary standard solution in IUPAC terminology.[ref]
standard solution. IUPAC Compendium of Chemical Terminology, fifth edition (Gold Book) by International Union of Pure and Applied Chemistry; Gold Book, standard solution, S05924; definitions of primary standard and secondary standard solution; underlying source Orange Book, second edition, p. 48
An accurate balance determines total mass, including any unwanted water or other material. To infer reacting inventory, one also needs known composition, stability during storage and weighing, complete transfer and dissolution, and a sufficiently complete chemical reaction of known stoichiometry. A finely resolved balance cannot establish these chemical facts. Potassium hydrogen phthalate is useful because a suitably prepared portion provides a known acid inventory for sodium hydroxide titrant standardization; the KHP standardization protocol below shows how that claim is put into practice.
Preparation belongs to the particular material and method. For example, the MIT handout specifies drying potassium hydrogen phthalate at for hours or overnight, cooling in a desiccator, and weighing by difference; its footnote identifies removal of superficial moisture as the purpose. This is a condition of that potassium hydrogen phthalate procedure, not a universal instruction to heat every primary standard in the same way.[ref]

Let be the transferred sample mass, the known mass fraction of the intended chemical compound, and its molar mass. In a model with complete dissolution and chemically inactive impurities,The purity fraction is dimensionless: would mean , not . These equations assume that the impurity contributes no titrant demand; an impurity that also reacts needs its own inventory equation. The value of must refer to the same preparation state as , for example the specified dry material. A mass fraction assigned before unaccounted uptake of water cannot simply be used after it.
For an original worked example, stipulate pure potassium hydrogen phthalate, abbreviated KHP, so , a selected mass , and rounded molar mass . The NIST Chemistry WebBook lists formula and molecular weight , consistent with the rounded molar mass used here.[ref]

The selected mass, purity assumption and volumes in this example are chosen inputs, not measured results or a claim about a real batch's purity. The acid-bearing anion of potassium hydrogen phthalate loses one proton to hydroxide. The net chemical equation isThus sodium hydroxide has one-to-one stoichiometry with potassium hydrogen phthalate in this neutralization, givingChoose a delivered endpoint titre of and matched titration blank of . . Original calculated inventory diagram for selected potassium hydrogen phthalate, stipulated , rounded molar mass , and one-to-one sodium hydroxide neutralization. The endpoint and matched titration blank are chosen examples, not observations. Details. When the entire weighed portion is titrated, added water changes concentration and the pH path but preserves the initial acid inventory. The dissolution volume therefore does not appear in . If instead a volumetric flask prepares a stock and a volumetric pipette selects an aliquot, the selected inventory is for homogeneous stock, with delivered aliquot volume and final stock volume . The same bookkeeping explains why complete transfer matters even when the amount of dissolution water is approximate.
Quasistatic process 2026-10-14
A quasistatic process is an ideal limit in which a changing thermodynamic system passes through a sequence of states arbitrarily close to thermodynamic equilibrium. Its intermediate pressure, temperature and volume are therefore well defined and obey its equation of state. Driving must be slow compared with the system's own equilibration, rather than merely taking a long time on a clock. A reversible thermodynamic process additionally allows both system and surroundings to be restored without net change; friction can produce entropy even in a quasistatic process.[ref]
For a computed example, take a fixed amount of substance of ideal gas in a sealed cylinder with a frictionless piston, in contact with a heat bath at absolute temperature . Reduce the external load in infinitesimal steps so that the piston expands from to . In the reversible limit, the gas pressure exceeds the external pressure by an infinitesimal amount during expansion, and the heat transfer occurs across an infinitesimal temperature difference. Exact mechanical equilibrium would give no driving force; the limiting imbalance supplies motion while preserving equilibrium intermediate states.[ref]
Along this reversible isothermal expansion, the ideal gas law givesHere is the molar gas constant; specifying fixes an energy scale, without choosing and separately. The initial and final equilibrium states are and . Count work delivered to the surroundings as positive. The reversible work is the shaded area beneath the pressure-volume curve:The integral of supplies the natural logarithm. Since internal energy of an ideal gas depends only on temperature, ; the first law of thermodynamics, , gives to the displayed precision. The heat bath replaces the energy delivered as work.[ref]
Now start from the same initial state, but abruptly lower the external pressure to the constant value . There is a finite initial pressure imbalance, so this is an irreversible expansion rather than a quasistatic process. Assume the piston begins and ends at rest, has no net change in stored energy, and has no external friction; relaxation occurs within the gas and its heat bath. After motion has settled and the heat bath has restored temperature , mechanical equilibrium and the ideal gas law give . The work delivered to the constant load isThis work follows directly from load force times piston displacement, using for piston area . Intermediate gas states need not have a single equilibrium pressure or temperature. In particular, neither at the bath temperature nor describes the actual gas during this irreversible motion. The rectangle below plots the known external load; its upper edge is not an equilibrium gas path.
The same equilibrium endpoints give different work for reversible and constant-load expansion
. The blue curve is the reversible ideal gas path. The dashed orange line is constant external pressure, whose rectangle gives of load work. The additional blue shading is available in the reversible case. Black points mark the common gas endpoints; the open orange point marks the initial external load, and the dotted gap indicates the initial imbalance. No irreversible internal gas path is drawn. Details. The entropy accounting makes the difference precise. Because entropy is a state function, both expansions have the same gas entropy change, computed along the reversible isothermal expansion:The thermal reservoir stays at and loses heat , so . Assume the ideal load stores work without an entropy change. The combined entropy production is thereforeNo numerical entropy unit is implied by alone: or must also be specified. The lost work opportunity is for this comparison. This calculation uses equilibrium endpoints and the reservoir's temperature, without assigning equilibrium intermediate states to the irreversible gas. [ref]
https://openstax.org/books/university-physics-volume-2/pages/4-6-entropy. OpenStax, §4.6: entropy as a state function and reversible reference paths
For another boundary choice, let the same ideal gas expand into an initially evacuated compartment of volume inside a rigid, insulated vessel of total volume . Removing the partition causes free expansion: no load is moved and no heat crosses the outer boundary, so and . After equilibration the temperature is again , the pressure is , and the gas entropy has increased by . The outer vessel has fixed volume; the gas's accessible volume doubles. Restoring the initial gas state requires compression and removal of heat, changing the surroundings. Equal endpoints do not make free expansion reversible. [ref]
https://openstax.org/books/university-physics-volume-2/pages/4-1-reversible-and-irreversible-processes. OpenStax, §4.1: insulated free expansion and restoration
A slowly moving piston with dry friction supplies a different caution: its gas can remain arbitrarily close to thermodynamic equilibrium, yet rubbing still converts work into heat and produces entropy. If the frictional force has a nonzero slow-motion limit, its dissipated energy over a fixed travel does not vanish with speed. Reversing the motion reverses friction, so infinitesimally changing the external load cannot retrace the same operation and restore the surroundings. The heat capacity calculations additionally exclude friction and other dissipative work.[ref]
The usefulness of a quasistatic process is that it replaces detailed motion by a readable sequence of equilibrium states. An equation of state and an integral then turn a pressure-volume drawing into an energy budget. The beautiful distinction is that internal energy and entropy depend on the endpoints, while heat and work record how the change was performed. Reversible benchmarks expose how much work an irreversible device sacrifices, while entropy production explains why simply making it slower need not recover that opportunity.
Racemate 2026-10-14
A racemate contains the same amount of substance of each member of an enantiomer pair. Their equal and opposite contributions cancel in ordinary solution polarimetry under identical conditions. Zero net optical rotation therefore does not establish achirality of the individual molecules.[ref]

Reaction rate 2026-10-13
For one chemical reaction in a closed system of fixed volume , with uniform concentrations and no competing reactions or appreciable accumulation of unlisted species, choose the chemical equationBrackets denote amount of substance per volume, for example . The stoichiometry gives one instantaneous reaction ratewhere is the existing extent of reaction. The minus signs make forward consumption positive; dividing by the coefficients makes measurements of different chemical species agree. Units can be . See IUPAC Gold Book: rate of reaction and Flowers et al., Chemistry 2e, §12.1: chemical reaction rates.
For the illustrative chemical equation , a measured implies , , and . These numbers express stoichiometry, without supplying a rate law.
Each derivative is the slope of a concentration curve at one instant. Over a finite interval, instead gives an average reaction rate. For A converting reversibly to B, the net reaction rate is ; both directional reaction rates can remain positive when at chemical equilibrium.
Redox titration Created 2026-10-13 Updated 2026-10-20
A redox titration adds a measured amount of substance of one reactant to determine another through a known redox chemical equation. The burette reading becomes an amount through concentration; the balanced chemical equation supplies the amount ratio. A colour endpoint additionally needs a chemical explanation, as in the permanganate-iron titration.
The useful bridge is an electron inventory. If each sample entity supplies electrons and each titrant entity accepts , matching the formal exchanged amount of substance of electrons givesHere is the titrant molar concentration and the volume attributable to the sample at the equivalence point. These are net half-reaction counts, not a claim that free electrons accumulate in the flask. The redox conversion must be sufficiently complete and selective for the stated stoichiometry to identify .
This is why a small visible signal can reveal a much larger invisible inventory: the signal locates a stopping point, while titrant standardization and stoichiometry turn delivery into amount of substance. The titration endpoint estimates the equivalence point; repeatable colour alone establishes neither correct titrant concentration nor chemical selectivity. The permanganate-iron endpoint and blank model separates these assumptions.
Repeated-bath dialysis model 2026-10-14
Consider an original ideal model with sample volume , bath volume , and initial sample solute concentration . Assume each volume is fixed, a freely permeant, nonbinding solute, no chemical reactions, complete mixing on each side, and equal concentrations at equilibrium. Each fresh bath initially contains none of this solute. The protein is perfectly retained in this model.
The first equilibration redistributes the initial amount of substance through both compartments. Conservation givesThis denominator includes the sample itself. For the chosen and , , rather than . Nine-tenths of the initial solute is in the bath and one-tenth remains in the sample; nothing has vanished during equilibration.
Discarding the equilibrated bath removes its solute. A second fresh bath starts with only of solute in the entire system, so the same argument applies again. Induction gives the geometric progressionHere counts completed equilibrations with fresh baths: is the first bath, follows one replacement, and follows two replacements. The normalized sample concentrations are respectively , and . Leaving the first bath in place for longer cannot lower the ideal equilibrium value below .
Repeated fresh baths reduce normalized solute concentration geometrically
. Original fixed-volume model, not experimental data. The left panel normalizes the first bath's conserved amount of substance by , distinguishing the sample's share from the bath's share. The right panel plots sample solute concentration divided by after each completed fresh-bath equilibration on a logarithmic vertical axis. The protein curve is normalized independently by its own initial concentration. The perfectly retained protein stays at normalized concentration one only because its amount of substance and the sample volume are fixed. Details. The same conservation argument handles a fresh bath already containing a permeant solute at fixed concentration :Thus the sample approaches , not necessarily zero. For example, if a hypothetical new buffer component starts at zero in the sample and at in every bath, its sample concentration reaches , and in the three equilibrations. Outward removal and inward replacement follow the same mathematics.
Equilibrium fractions do not specify elapsed time. An illustrative well-mixed linear ordinary differential equation model within one bath isHere is an effective transfer coefficient with units of volume per time. Adding the two equations conserves ; subtracting their normalized forms givesThe difference undergoes exponential decay. A bath replacement resets , renewing the driving difference without making jump instantly. The plotted points are equilibrium endpoints, not a measured time course or a timetable for an arbitrary protein.
Real sample volume can change through osmosis; proteins can be lost through leakage or adsorption to the semipermeable membrane. Consequently, a measured decrease in protein concentration alone cannot distinguish dilution from loss of protein. Measuring recovered volume as well as protein concentration permits an estimate of recovered protein mass.[ref]

Repeated ideal-stage enrichment 2026-10-18
Stipulate a constant relative volatility . The vapor-liquid equilibrium model then becomesAn initial liquid mole fraction gives an equilibrium vapor mole fraction . Now imagine completely condensing a small sample of that vapor and using this condensate as a fresh liquid for another equilibration. Repeating this formal operation givesThese are original model calculations of ideal-stage enrichment. They are not measured compositions, a reflux design, or successive compositions of one pot undergoing simple distillation. That pot loses the more volatile component and becomes depleted; each step here instead starts from the previous enriched condensate. The displayed sequence specifies compositions, not the recoverable amount at each step.
Respiratory control ratio 2026-10-18
The respiratory control ratio divides the oxygen consumption rate during ADP-stimulated respiration by that after supplied ADP has been used. It compares reaction rates, not ATP formed per oxygen.[ref]
Respiratory enzymes in oxidative phosphorylation. I. Kinetics of oxygen utilization by Britton Chance and G. R. Williams. pp. 386–387, Figure 2 and discussion of the ratio of respiration rates with and without ADP
A large ratio means strong stimulation under the comparison's conditions; it is not a universal measure of all mitochondrial functions.
For a constructed example, choose constant suspension volume and oxygen concentration declines of and during the two comparison windows. Assume no gas exchange or dilution and a correctly calibrated readout. A derivative converts each concentration slope into an amount of substance consumption rate:The common volume cancels in the ratio, so dividing the positive slope magnitudes gives the same answer. These are chosen inputs, not the 1955 observations. If an uncorrected influx of of oxygen occurred in both windows, the apparent rates would instead be and , giving . The same absolute error can inflate a ratio because it removes a larger fraction of its smaller denominator.
Respirometer gas balance 2026-10-14
For an original idealized mathematical model, let be the amount of substance of molecular oxygen consumed since the initial reading and the carbon dioxide produced. Suppose a fraction of that carbon dioxide is removed from the gas, all other gas inventories remain fixed, and temperature and gas pressure stay constant. The ideal gas law gives the positive gas contractionwhere is the molar gas constant and the gas volume. With complete absorption, , the contraction reflects molecular oxygen uptake regardless of the amount of carbon dioxide made. For a uniform pipette bore of area , an inward boundary displacement gives . A graduated pipette can instead supply the displaced volume directly.
Let denote sample contraction and the matched blank contraction. Define with both taken relative to their own initial readings. Under the preceding assumptions,The slope of corrected volume against time estimates uptake over the chosen interval. The approximate sign acknowledges that a liquid boundary also responds to hydrostatic and capillary pressure, while the gas can exchange water vapour. A fixed-volume apparatus would instead show a pressure decrease; its ideal conversion is after complete absorption and appropriate blank correction.
The ideal gas law also exposes drift without any biology: for fixed gas inventory, . A one-percent rise in absolute temperature at fixed pressure produces a one-percent expansion. Matching the initial gas spaces makes such common changes easier to subtract. Subtracting unmatched blanks can leave a physical signal that looks biological.
Saponification with limiting hydroxide 2026-10-18
Consider a hypothetical closed, well mixed chemical solution initially containing of ethyl acetate, of hydroxide, and no acetate ions or ethanol. One millimole is mole. Assume complete conversion as far as the one-to-one chemical equation allows, with no competing chemical reactions. This is an ideal stoichiometry calculation, not a measured yield.
Let the extent of reaction be , expressed in millimoles. The four values of amount of substance, in the order ethyl acetate, hydroxide, acetate ion, ethanol, areThe upper bound follows because both reactant quantities must be nonnegative. Thus hydroxide is the limiting reagent, and the ideal endpoint leavesThe carboxylate formed equals the hydroxide consumed, while unreacted ester remains. Adding more hydroxide raises the stoichiometry bound; merely waiting longer does not change that bound. To convert these values of amount of substance into concentrations, a volume is also required. The endpoint does not specify the reaction rate or time needed to approach it.
SEC partition coefficient 2026-10-14
For ideal SEC, define the SEC partition coefficient byHere averages solute concentration over all pore liquid, including inaccessible regions. is column elution volume. Pure steric exclusion gives ; the endpoints mean exclusion and full access.[ref]

A storage argument explains the relation. Assume a dilute stable solute, unchanged packing and solvent, constant volumetric flow rate , rapid local chemical equilibrium between pore liquid and interparticle liquid, negligible binding, and negligible transport through the pores by bulk flow. At interparticle concentration , the column holds an amount of substance . Its exit transports per unit time. The ratio is the mean residence time in this equilibrium transport model; multiplying by gives . For a narrow symmetric SEC chromatogram peak, the peak maximum approximates that mean; a strongly asymmetric peak needs an explicitly stated location convention.
The handbook also uses an operational normalizationThis denominator includes solid matrix, so full access gives when . It differs from the pore-liquid SEC partition coefficient. Read each source's volume convention before comparing coefficients.[ref]

Selective biochemical reaction pathways 2026-10-14
An enzyme accelerates particular chemical reactions, with catalytic selectivity arising from recognition and from the relative ease of the subsequent chemistry. Strong binding alone does not guarantee fast conversion. Nor does catalysis change the chemical equilibrium for an unchanged net chemical reaction. These distinctions explain how enzymes can direct the flow of material without creating an extra source of free energy.[ref]

For an original model of competing routes, let a substrate become either a desired reaction product or another reaction product :The labels represent hypothetical chemical species with the same elemental composition, not molecular structures. Assume fixed volume, irreversible first-order reactions, constant effective rate constants, and initially of with neither reaction product present. The model approximates a limited operating regime; real enzyme kinetics can include saturation and reverse reactions.
Selective acceleration changes the split between competing biochemical reaction products
. Original illustrative chemical reaction network and limiting product amounts from of hypothetical . Chosen effective rate constants are or and . The boxes label chemical species; they are not molecular structures. The bars are model limits, not experimental data. Details. The reaction-rate equations give , and , where denotes amount of substance. Consequently,With and , the eventual split is of and of . At ten minutes, the calculated amounts are approximately , and for , and . Selectively increasing fourfold changes the eventual desired fraction to . Increasing both rate constants fourfold instead preserves the split and only shortens the conversion time. Speed and selectivity answer different questions.
The bookkeeping matters independently of that model. In ideal alcoholic fermentation, complete conversion of of glucose yields each of ethanol and carbon dioxide. Of the initial of carbon chemical atoms, remain in ethanol and enter carbon dioxide. This is a chosen material-balance example using the net stoichiometry explained under Saccharomyces cerevisiae; it gives neither an observed yield nor the mechanism of the individual steps. Equal amounts of the two reaction products also do not mean equal masses.
Solubility product and material balance 2026-10-18
A solubility product constrains a product of thermodynamic activities; the available inventories determine which combinations of concentrations are possible. Their intersection determines the molar solubility. Consider an original, abstract AB solid, with the preceding one-to-one dissolution chemical equation. Stipulate , and at fixed temperature and pressure. Thus is dimensionless. These are chosen model parameters, not data for an identified material.
Initially, a completely dissociated soluble XB salt supplies B anions at molar concentration and X cations at the same molar concentration; A is absent. All three ions have unit electric charge magnitude. X remains a spectator ion, and only AB can form a solid. Assume constant volume, no additional chemical reactions, and negligible contributions from self-ionization of water. Dissolving AB raises both reacting-ion concentrations by the same :Charge neutrality is satisfied: . Adding B alone without its accompanying cation would omit the initial electric charge balance.
In the plane of the two reacting-ion concentrations, this inventory constraint is the line . Saturation supplies the hyperbola for . Their positive intersection solves a quadratic equation:The quadratic formula supplies two roots of a polynomial; only the positive one is a physical concentration. Uniqueness also follows because strictly increases for , starting at zero and growing without bound. The second expression, obtained by multiplying by the conjugate, avoids subtraction of nearly equal numbers at large .

Intersections of the saturation hyperbola with inventory lines give the pure-water and common-ion chemical equilibrium compositions. The dashed projections read the intersection; these curves are calculated constraints, not measured data or a time evolution. Details. For pure water, gives . For ,Their product is , although neither concentration equals the pure-water value. This is the useful geometrical distinction: an inventory line and an equilibrium constant specify different constraints.
Check the solid inventory before imposing saturation. If the initially added AB has amount of substance in volume , the saturated branch requires for excess solid to remain. For example, choose and . In pure water, dissolves and the same amount of substance remains as solid. At , approximately dissolves, leaving as solid. In this model, an insufficient supply instead givesIf , all AB dissolves and the ion product is below the solubility product. At , the last solid disappears exactly at saturation.

How can an invisible dissolved inventory be measured? The unsigned laboratory handout Solubility Product Constant, Ksp, for Calcium Hydroxide by Acid-Base Titration, Procedure, starts with saturated calcium hydroxide, supplied by the teacher or prepared from calcium and water. Its apparatus includes filter paper, a funnel, stoppered flasks, a volumetric pipette, a volumetric flask and a burette. Two successive rounds of filtration remove particles. The filtrate is stoppered to reduce carbon dioxide exposure. A aliquot is diluted with deionized water and titrated with standardized hydrochloric acid; bromothymol blue gives a pale-green titration endpoint. The readout is the difference between final and initial burette readings; four consistent trials are required.[ref]
Unlike AB, calcium hydroxide has one-to-two dissolution stoichiometry:For molar solubility in the ideal model with no background calcium or hydroxide, no additional chemical reactions, and negligible self-ionization of water, and . Hence , with units ; it is not the AB constant used in the figures. The neutralization chemical equation is . Let be the known titrant molar concentration, its delivered volume at the equivalence point, and the original aliquot volume. Then the inventory calculation isDilution of the aliquot changes its concentration during the acid-base titration, but preserves its initial amount of substance of hydroxide. Therefore , rather than the diluted flask volume, belongs in the denominator.
The handout identifies two biases: carried-over calcium hydroxide solid dissolves during acid-base titration, consuming extra titrant; carbon dioxide consumes hydroxide and forms calcium carbonate. Filtration and stoppering address these effects, while repeat trials test consistency. Its review questions also ask about dissolved ion pairs, which filtration cannot remove.[ref]
Further controls would compare a water blank, independently verify titrant standardization, and record the saturation temperature; these are proposed checks, not reported measurements. A titration endpoint estimates the equivalence point and may introduce a systematic offset. Even perfect inventory measurement does not determine activity coefficients. Recovering the dimensionless thermodynamic solubility product additionally requiresThis explains both the power and the limit of the experiment: acid-base titration measures a dissolved inventory, while the link from that inventory to a thermodynamic equilibrium constant still depends on a chemical model.
